Anti-Piezo1 (extracellular) Antibody (#APC-087) is a highly specific antibody directed against an extracellular epitope of the rat protein. The antibody can be used in western blot, immunohistochemistry and live cell flow cytometry applications. It has been designed to recognize Piezo1 from mouse and rat samples. The antibody is will not recognize the protein from human samples. Anti-Piezo1 (extracellular)-APC Antibody (#APC-087-APC) is directly conjugated to the Allophycocyanin (APC) fluorophore. This conjugated antibody has been developed to be used in immunofluorescent applications such as direct flow cytometry and live cell imaging. It has been designed to recognize Piezo1 from mouse and rat samples.

Target PIEZO1

Overview

Anti-Piezo1 (extracellular)-APC Antibody is an antibody targeting Piezo-type mechanosensitive ion channel component 1, Protein FAM38A, Mib, Membrane protein induced by beta-amyloid treatment Polyclonal raised in Rabbit (Allophycocyanin (APC)). This antibody is commonly used in FC, LCI to detect, localize, or compare expression of the target across samples.

Key elements and design rationale

  • Target: Piezo-type mechanosensitive ion channel component 1, Protein FAM38A, Mib, Membrane protein induced by beta-amyloid treatment (also reported as Piezo-type mechanosensitive ion channel component 1, Protein FAM38A, Mib, Membrane protein induced by beta-amyloid treatment).
  • Immunogen/epitope region: The peptide is located in an internal region of the rat Piezo1 (the exact topology of Piezo1 remains to be defined)..
  • Homology note: Mouse - 12 out of 14 amino acids residues identical It won't recognize human Piezo1 (informative for cross-species interpretation).
  • Species reactivity (as provided): Rat, Mouse.
  • Lot quality control (as provided): Western blot analysis (unlabeled antibody, #APC-087), and direct flow cytometry (labeled antibody)..
  • Peptide confirmation: Confirmed by amino acid analysis and mass spectrometry.
  • Blocking peptide: Available for antigen preadsorption control where appropriate.
  • Conjugate/format: Allophycocyanin (APC) (may affect detection channel and background).

These attributes help researchers interpret whether signal reflects the intended target in a given assay and sample context.

Biological background

Piezo1 protein is a family member of mixed cationic, non selective, directly mechanically activated ion channels found in higher eukaryotic cells. Piezo proteins are associated with physiological responses in cells that have been identified in many cell types and tissues in several species where mechanosensitivity plays a major role1,2.Two mammalian Piezo proteins were identified, piezo1 and piezo2 encoded by the fam38A and fam38B genes recpectively2. Piezo1 expression is detected in organs such as: bladder, colon, kidney, lung and skin3.The secondary structure and general length of Piezo proteins are quite conserved among species, and seem to form homotetrameric aggregates.

Research relevance and current trends

  • Linking transporter/channel abundance to ionic homeostasis and excitability-related phenotypes.
  • Studying compartment-specific localization (surface vs intracellular pools) and trafficking dynamics.
  • Combining antibody readouts with functional assays for more complete interpretation.

Common research applications

  • Flow cytometry (direct/indirect): quantify target-positive populations and shifts in expression across subsets.
  • Live cell imaging (LCI): support extracellular-epitope detection on non-permeabilized cells when appropriate.

Interpretation typically benefits from comparing matched sample sets (e.g., treated vs control, WT vs KO/KD) and using orthogonal readouts where feasible.

Notes for experimental interpretation

  • Isoforms and post-translational modifications can shift apparent molecular weight or epitope accessibility across samples.
  • Cross-species signal may depend on epitope conservation; consult the provided homology note when selecting models.
  • Permeabilization, fixation, and antigen retrieval can change accessibility of intracellular vs extracellular epitopes.
  • Conceptual control: antigen preadsorption (blocking peptide) can help assess signal dependence on the immunogen region.
  • Provided control suggestions: Negative control: RIC-001-APC.
  • Application notes: see product-specific dilution/usage notes and control concepts provided in the dataset.
