Product Overview
This rabbit polyclonal antibody detects phosphoinositide 3-kinase regulatory subunit 4 (gene PIK3R4) in human samples and is validated for Western blot, flow cytometry and ELISA. The predicted molecular weight is 153.1 kDa, and the supplier reports an observed band at about 150 kDa.
PIK3R4 (VPS15) is the regulatory partner of the class III PI3K VPS34 and forms the core of complexes that produce PI3P. These complexes drive autophagosome formation, endosome maturation and endocytic sorting, so VPS15 is studied alongside VPS34 and Beclin 1 in autophagy and membrane trafficking.

Validated Applications
| Western blot | 0.25–0.5 µg/mL (Human) |
|---|---|
| Flow cytometry (fixed cells) | 1–3 µg/1×106 cells (Human) |
| ELISA | 0.1–0.5 µg/mL |
Samples with a confirmed band (WB): human HeLa cells, human HepG2 cells, human 293T cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with a confirmed signal (flow cytometry): HepG2 cells.
Immunogen
Recombinant human PIK3R4 fragment (Arg36–Gln1339), expressed in E. coli. Sequence identity with the mouse and rat orthologs: 95.7% and 95.6%, respectively.
Reactivity Notes
The supplier lists reactivity with human. UniProt places phosphoinositide 3-kinase regulatory subunit 4 in the late endosome, cytoplasmic vesicle and membrane. Tissue expression noted by UniProt (human): Ubiquitously expressed. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
