Product Overview
This rabbit polyclonal antibody recognizes pirin in human, monkey, mouse and rat samples and is validated for Western blot, ICC/IF, flow cytometry and ELISA. The predicted molecular weight is 32.1 kDa, and the supplier reports an observed band at about 36 kDa.
Pirin is a nuclear iron-containing protein of the cupin family that acts as a redox-sensitive coregulator of NF-κB, helping NF-κB proteins bind target DNA. It also shows quercetin dioxygenase activity in vitro and is studied in oxidative stress responses, myeloid differentiation and cancer cell migration.

Validated Applications
| Western blot | 0.1–0.25 µg/mL (Human, Monkey, Mouse, Rat) |
|---|---|
| Immunocytochemistry / Immunofluorescence | 5 µg/mL (Human) |
| Flow cytometry (fixed cells) | 1–3 µg/1×106 cells (Human) |
| ELISA | 0.1–0.5 µg/mL |
Samples with a confirmed band (WB): human HepG2 cells, human HeLa cells, monkey COS-7 cells, rat heart tissue, rat RH-35 cells, mouse NIH/3T3 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.25 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (ICC/IF): U2OS cells.
Samples with a confirmed signal (flow cytometry): HepG2 cells.
Immunogen
Recombinant human PIR fragment (Lys5–Gln266), expressed in E. coli. Sequence identity with the mouse and rat orthologs: 96.6% and 95.8%, respectively.
Reactivity Notes
The supplier lists reactivity with human, monkey, mouse and rat. UniProt places pirin in the nucleus and cytoplasm. Tissue expression noted by UniProt (human): Highly expressed in a subset of melanomas. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
