Product Overview
Clone CHO-16 is a rabbit monoclonal antibody that recognizes protein kinase C alpha type (gene PRKCA) in human, mouse and rat samples and is validated for Western blot, IHC, ICC/IF, immunoprecipitation and flow cytometry. The predicted molecular weight is 76.8 kDa, and the supplier reports an observed band at about 77 kDa.
PKCα (PRKCA) is a conventional protein kinase C isoform activated by calcium, diacylglycerol and phospholipids. It phosphorylates targets such as RAF1 and BCL2 and feeds into ERK/MAPK signaling, influencing cell proliferation, survival, adhesion and migration, and it is studied in cancer, cardiac function and platelet biology.

Samples with a confirmed band (WB): human HeLa cells, human 293T cells, human Jurkat cells, human A549 cells, rat liver tissue, rat spleen tissue, mouse liver tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:1000 overnight at 4 °C, HRP-conjugated secondary antibody at 1:500, ECL detection.
Samples with confirmed staining (IHC): human pituitary tumor tissue, human melanoma tissue, human thymoma tissue, human colon tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Immunogen
Synthetic peptide based on the sequence of human PKC alpha.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places protein kinase C alpha type in the cytoplasm, cell membrane, mitochondrion membrane and nucleus. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
