Product Overview
This rabbit polyclonal antibody recognizes plakophilin-2 (gene PKP2) in human, mouse and rat samples and is validated for Western blot, IHC, ICC/IF, flow cytometry and ELISA. The predicted molecular weight is 97.4 kDa, and the supplier reports an observed band at about 92 kDa.
Plakophilin-2 is an armadillo-repeat protein of desmosomes, the adhesive junctions that anchor intermediate filaments between neighboring cells. It is the main plakophilin of heart muscle, and PKP2 mutations are the most common genetic cause of arrhythmogenic right ventricular cardiomyopathy.

Validated Applications
| Western blot | 0.25–0.5 µg/mL (Human, Mouse, Rat) |
|---|---|
| IHC (paraffin sections) | 2–5 µg/mL (Human, Mouse) |
| Immunocytochemistry / Immunofluorescence | 5 µg/mL (Human) |
| Flow cytometry (fixed cells) | 1–3 µg/1×106 cells (Human) |

Samples with a confirmed band (WB): human Hacat cells, rat heart tissue, mouse heart tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human lung cancer tissue, human colon adenocarcinoma tissue, human liver cancer tissue, human tonsil tissue, human urothelial carcinoma tissue, mouse cardiac tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Samples with confirmed staining (ICC/IF): U2OS cells.
Samples with a confirmed signal (flow cytometry): U87 cells.
Immunogen
Recombinant human PKP2 fragment (Arg369–Gln804), expressed in E. coli. Sequence identity with the mouse and rat orthologs: 82.8% and 79.4%, respectively.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places plakophilin-2 in the nucleus, cell junction and cytoplasm. Tissue expression noted by UniProt (human): Expressed at myocyte intercalated disks in the heart (at protein level). The samples tested by the supplier (listed above) are a practical starting point for positive controls.
