Product Overview

This rabbit polyclonal antibody recognizes group 3 secretory phospholipase A2 (gene PLA2G3) in human and mouse samples and is validated for Western blot, flow cytometry and ELISA. The predicted molecular weight is 57.2 kDa, and the supplier reports an observed band at about 65 kDa.

Group III secreted phospholipase A2 is a Ca2+-dependent enzyme related to bee venom PLA2. It hydrolyzes membrane and lipoprotein phospholipids and has been studied in lipid mediator production, sperm maturation, mast cell function and lipoprotein remodeling.

Validated Applications

Western blot 0.25–0.5 µg/mL (Human, Mouse)
Flow cytometry (fixed cells) 1–3 µg/1×106 cells (Human)
ELISA 0.1–0.5 µg/mL

Samples with a confirmed band (WB): human K562 cells, mouse testis tissue.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.

Samples with a confirmed signal (flow cytometry): HepG2 cells.

Immunogen

Recombinant human PLA2G3 fragment (Glu74–Arg358), expressed in E. coli. Sequence identity with the mouse ortholog: 66.8%.

Reactivity Notes

The supplier lists reactivity with human and mouse. UniProt describes group 3 secretory phospholipase A2 as secreted and also located in the cell membrane and recycling endosome. Tissue expression noted by UniProt (human): Expressed in kidney, heart, liver, and skeletal muscle. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

For research use only; not for diagnostic or therapeutic procedures. Handle with standard laboratory precautions.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 0.25–0.5 µg/mL (Human, Mouse); Flow cytometry (fixed cells) 1–3 µg/1×10⁶ cells (Human); ELISA 0.1–0.5 µg/mL. Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human, Mouse. Other species are not listed by the supplier.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 57.2 kDa; observed band at about 65 kDa. A difference from the calculated size is common and can reflect post-translational modification, processing or isoform differences.
Q.What immunogen was used to raise this antibody?
A.Recombinant human PLA2G3 fragment (Glu74–Arg358), expressed in E. coli. Sequence identity with the mouse ortholog: 66.8%.
Q.How should this antibody be stored?
A.As supplied: −20 °C for up to 12 months from receipt. After reconstitution: 4 °C for up to 1 month, or aliquot and keep at −20 °C for up to 6 months. Avoid repeated freeze–thaw cycles.
