Product Overview

Clone AFE-16 is a rabbit monoclonal antibody that recognizes plectin in human, mouse and rat samples and is validated for Western blot, IHC and immunoprecipitation. The predicted molecular weight is 531.8 kDa, and the supplier reports an observed band at about 531 kDa.

Plectin is a very large cytolinker protein that connects intermediate filaments to actin, microtubules and cell junctions such as hemidesmosomes and desmosomes. It gives mechanical strength to skin, muscle and other tissues, and PLEC mutations cause epidermolysis bullosa simplex with muscular dystrophy and related disorders.

Validated Applications

Western blot 1:500–1:2000
Immunohistochemistry 1:50–1:200
Immunoprecipitation 1:30

Samples with a confirmed band (WB): Jurkat cells.

Recommended loading (WB): 20–40 µg of total protein per lane.

Immunogen

Synthetic peptide based on the sequence of human plectin.

Reactivity Notes

The supplier lists reactivity with human, mouse and rat. UniProt places plectin in the cytoplasm, cell junction and cell projection. Tissue expression noted by UniProt (human): Widely expressed with highest levels in muscle, heart, placenta and spinal cord. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
