Key elements and design rationale

  • Target: PLXNA1 (Type II inositol 3,4-bisphosphate 4-phosphatase).
  • Antibody format: Monoclonal; clone AFOO-16; isotype Rabbit IgG.
  • Host: Rabbit.
  • Species reactivity: Human,Mouse,Rat (confirm in your model system with appropriate controls).

This description is intended to help interpret the antibody design and the biological context of the target using the fields provided in the catalog record, alongside general experimental considerations.

Biological background

PLXNA1 (protein: T-cell surface glycoprotein CD3 zeta chain) is a commonly studied target in molecular and cellular biology. Functional context (as provided): Catalyzes the hydrolysis of the 4-position phosphate of phosphatidylinositol 3,4-bisphosphate, inositol 1,3,4- trisphosphate and inositol 1,4-bisphosphate. Reported cellular localization context: Cytoplasm, cytoskeleton, microtubule organizing center, centrosome . Tissue expression notes (as provided): Widely expressed with highest levels occurring in the skeletal muscle and heart. .

Research relevance and current trends

  • Research context keywords from the source record include: Axonal Guidance Proteins,Growth and Development,More Gpcr,Neurogenesis,Neurology Process,Neuroscience,Neurotransmission,Receptors / Channels.
  • Current studies often focus on connecting target abundance/localization to pathway perturbations across models, tissues, and cell states.
  • Quantitative and multiplexed assays (e.g., imaging + immunoblot panels) are commonly used to compare phenotypes across conditions and time-courses.

Common research applications

  • Western blotting (WB): assess relative target abundance across samples, treatments, or time-points.
  • Immunofluorescence/ICC (IF/ICC): visualize subcellular localization patterns and cell-to-cell heterogeneity.
  • Flow cytometry: quantify target-positive populations and compare shifts in marker distributions.

Workflow ideas (metafield): Validate PLXNA1 antibody specificity using KO/KD control samples (WB/IF/IHC as appropriate), Detect PLXNA1 expression by Western blot in cell or tissue lysates, Localize PLXNA1 by immunofluorescence/immunocytochemistry in cultured cells, Quantify PLXNA1-positive cells by flow cytometry in single-cell suspensions

Notes for experimental interpretation

  • Consider isoforms and post-translational modifications (PTMs) that may shift apparent molecular weight or epitope accessibility.
  • Apparent molecular weight may vary by sample type and processing (observed MW: 72 kDa; calculated MW: 104738 MW).
  • Control concepts: include appropriate negative controls (e.g., isotype, KO/KD samples) and orthogonal validation when feasible.

Additional product details (from the source record)

  • Molecular weight (observed): 72 kDa
  • Cellular localization (provided): Cytoplasm, cytoskeleton, microtubule organizing center, centrosome .
  • Tissue details (provided): Widely expressed with highest levels occurring in the skeletal muscle and heart. .

Will M06377 anti-Plexin A1 Monoclonal antibody work on parafin embedded sections? If so, which fixation method do you recommend we use (PFA, paraformaldehyde, other)?
It shows on the product datasheet, M06377 anti-Plexin A1 Monoclonal antibody as been validated on IF. It is best to use PFA for fixation because it has better tissue penetration ability. PFA needs to be prepared fresh before use. Long term stored PFA turns into formalin, as the PFA molecules congregate and become formalin.
