Product Overview
This rabbit polyclonal antibody targets protein O-glucosyltransferase 1 (gene POGLUT1) in human and mouse samples and is validated for Western blot, IHC and ICC/IF. The predicted molecular weight is 46.2 kDa, and the supplier reports an observed band at about 55 kDa.
Protein O-glucosyltransferase 1 (POGLUT1) adds glucose or xylose to specific serine residues within EGF-like repeats of secreted and membrane proteins. Its best-known target is the Notch receptor, and this modification supports normal Notch signaling and muscle development. POGLUT1 mutations cause a form of limb-girdle muscular dystrophy.

Samples with a confirmed band (WB): human HL-60 cells, human HepG2 cells, human U87 cells, human Raji cells, human Jurkat cells, human HeLa cells, mouse thymus tissue, mouse Ana-1 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human breast cancer tissue, human gallbladder adenocarcinoma tissue, human gastric cancer tissue, human lung cancer tissue, human lymphoma tissue, human ovarian cancer tissue, human placenta tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Samples with confirmed staining (ICC/IF): T-47D cells.
Immunogen
Synthetic peptide based on the sequence of at C-terminus of human POGLUT1. Sequence identity with the mouse and rat orthologs: 92.6% and 88.9%, respectively.
Reactivity Notes
The supplier lists reactivity with human and mouse. UniProt places protein O-glucosyltransferase 1 in the endoplasmic reticulum lumen. Tissue expression noted by UniProt (human): Expressed in most adult tissues at different intensities. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
