Product Overview
Clone 21P14 is a rabbit monoclonal antibody that recognizes DNA polymerase delta catalytic subunit (gene POLD1) in human, mouse and rat samples and is validated for Western blot, IHC, ICC/IF and flow cytometry. The predicted molecular weight is 123.6 kDa, and the supplier reports an observed band at about 124 kDa.
POLD1 is the catalytic subunit of DNA polymerase δ, which carries out lagging-strand synthesis during DNA replication. It has both polymerase and 3′-to-5′ proofreading exonuclease activity, giving high copying fidelity. Polymerase δ also works in several DNA repair pathways, and POLD1 mutations are linked to cancer predisposition.

Samples with confirmed staining (IHC): human colon cancer, human lung adenocarcinoma tissue, human ovarian cancer tissue, human tonsil tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Immunogen
Synthetic peptide based on the sequence of human POLD1.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places DNA polymerase delta catalytic subunit in the nucleus. Tissue expression noted by UniProt (human): Widely expressed, with high levels of expression in heart and lung. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
