Product Overview
Clone 19P05 is a rabbit monoclonal antibody that recognizes serum paraoxonase/arylesterase 2 (gene PON2) in human, mouse and rat samples and is validated for Western blot, IHC and immunoprecipitation. The predicted molecular weight is 39.4 kDa, and the supplier reports an observed band at about 30 kDa.
Paraoxonase 2 (PON2) is an intracellular member of the paraoxonase family that is widely expressed but, unlike PON1, is not carried on HDL. It hydrolyzes certain lactones and shows antioxidant activity, reducing lipid peroxidation of LDL. PON2 is studied in oxidative stress, atherosclerosis and bacterial quorum-sensing signals.

Validated Applications
| Western blot | 1:1000–1:5000 |
|---|---|
| Immunohistochemistry | 1:50–1:200 |
| Immunoprecipitation | 1:50 |
Samples with a confirmed band (WB): human HeLa cells, human A549 cells, human U251 cells, human SiHa cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 10% SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:1000 overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human colon carcinoma tissue, human skin cancer tissue, human thyroid cancer tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Immunogen
Synthetic peptide based on the sequence of human PON2.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places serum paraoxonase/arylesterase 2 in the membrane. Tissue expression noted by UniProt (human): Widely expressed with highest expression in liver, lung, placenta, testis and heart. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
