Product Overview
This rabbit polyclonal antibody detects protection of telomeres protein 1 (gene POT1) in human, mouse and rat samples and is validated for flow cytometry and Western blot. The predicted molecular weight is 71.4 kDa, and the supplier reports an observed band at about 71 kDa.
Protection of telomeres 1 (POT1) is the shelterin component that binds the single-stranded TTAGGG overhang at chromosome ends. It shields telomeres from being recognized as DNA damage and helps control access of telomerase, thereby regulating telomere length. POT1 is studied in telomere biology, aging and cancer.

Validated Applications
| Western blot | 0.1–0.5 µg/mL (Human, Mouse, Rat) |
|---|---|
| Flow cytometry (fixed cells) | 1–3 µg/1×106 cells (Human) |
Samples with a confirmed band (WB): human HeLa cells, human K562 cells, rat brain tissue, rat liver tissue, mouse Neuro-2a cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 50 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:10000, ECL detection.
Samples with a confirmed signal (flow cytometry): U937 cells.
Immunogen
Synthetic peptide from the N-terminal region of human POT1. It differs from the mouse sequence at eleven residues.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places protection of telomeres protein 1 in the nucleus and chromosome. Tissue expression noted by UniProt (human): Ubiquitous. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
