Product Overview

This rabbit polyclonal antibody targets protein phosphatase 1D (gene PPM1D) in human, mouse and rat samples and is validated for Western blot, flow cytometry and ELISA. The predicted molecular weight is 66.7 kDa, and the supplier reports an observed band at about 67 kDa.

PPM1D, also called WIP1, is a p53-induced serine/threonine phosphatase that switches off the DNA damage response. It dephosphorylates p53, CHK1, p38 MAPK and other checkpoint proteins, allowing cells to resume cycling after repair. PPM1D is amplified or truncated in several cancers and is studied as a regulator of p53 signaling.

Samples with a confirmed band (WB): human HeLa cells, human MDA-MB-453 cells, human Jurkat cells, human 293T cells, rat kidney tissue, rat PC-12 cells, mouse kidney tissue.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.

Samples with a confirmed signal (flow cytometry): A431 cells.

Reactivity Notes

The supplier lists reactivity with human, mouse and rat. UniProt places protein phosphatase 1D in the nucleus and cytoplasm. Tissue expression noted by UniProt (human): Expressed in fetal and adult brain. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
