Product Overview
This rabbit polyclonal antibody recognizes protein arginine N-methyltransferase 1 (gene PRMT1) in human samples and is validated for Western blot, IHC, flow cytometry and ELISA. The predicted molecular weight is 42.5 kDa, and the supplier reports an observed band at about 42–45 kDa.
PRMT1 is the main type I protein arginine methyltransferase in mammalian cells, producing monomethyl and asymmetric dimethyl arginine. It deposits the activating histone mark H4R3me2a and methylates many RNA-binding and signaling proteins, influencing transcription, RNA processing and the DNA damage response, and it is studied in cancer and development.

|---|---|
| IHC (paraffin sections) | 0.5–1 µg/mL (Human) |
| Flow cytometry (fixed cells) | 1–3 µg/1×106 cells (Human) |
| ELISA | 0.1–0.5 µg/mL |
Samples with a confirmed band (WB): human HEK293 cells, human A549 cells, human Caco-2 cells, human PC-3 cells, human K562 cells, human HeLa cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 50 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:10000, ECL detection.
Samples with confirmed staining (IHC): human rectal cancer tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in citrate buffer (pH 6).
Samples with a confirmed signal (flow cytometry): 293T cells.
Immunogen
Recombinant human PRMT1 fragment (Met11–Lys145), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with human. UniProt places protein arginine N-methyltransferase 1 in the nucleus, cytoplasm and lysosome membrane. Tissue expression noted by UniProt (human): Widely expressed. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
