Product Overview

This rabbit polyclonal antibody recognizes prominin-1 (gene PROM1) in human samples and is validated for Western blot, ICC/IF, flow cytometry and ELISA. The predicted molecular weight is 97.2 kDa, and the supplier reports an observed band at about 97 kDa.

Prominin-1 (PROM1, CD133) is a pentaspan membrane glycoprotein concentrated in plasma membrane protrusions such as microvilli and cilia, where it binds cholesterol. It is a widely used surface marker of hematopoietic and neural stem cells and of cells with stem-like properties in several tumors, and it is required for photoreceptor disk formation.

Validated Applications

Western blot 0.25–0.5 µg/mL (Human)
Immunocytochemistry / Immunofluorescence 4 µg/mL (Human)
Flow cytometry (fixed cells) 1–3 µg/1×106 cells (Human)
ELISA 0.1–0.5 µg/mL

Samples with a confirmed band (WB): human placenta tissue, human Caco-2 cells.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.

Samples with confirmed staining (ICC/IF): A549 cells.

Samples with a confirmed signal (flow cytometry): Caco-2 cells.

Immunogen

Recombinant human PROM1 fragment (Asp27–Arg821), expressed in E. coli.

Reactivity Notes

The supplier lists reactivity with human. UniProt places prominin-1 in the apical cell membrane, cell projection and endoplasmic reticulum. Tissue expression noted by UniProt (human): Isoform 1 is selectively expressed on CD34 hematopoietic stem and progenitor cells in adult and fetal bone marrow, fetal liver, cord blood and adult peripheral blood. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
