Product Overview

Clone 4G13G9 is a mouse monoclonal antibody that recognizes prothrombin (gene F2) in human samples and is validated for Western blot and IHC. The predicted molecular weight is 70.0 kDa, and the supplier reports an observed band at about 70 kDa.

Prothrombin (F2) is the vitamin K-dependent precursor of thrombin, the central protease of blood coagulation. Thrombin converts fibrinogen to fibrin, activates factors V, VIII, XI and XIII and platelets through PAR receptors, and with thrombomodulin activates the anticoagulant protein C. The prothrombin G20210A variant is a well-known risk factor for venous thrombosis.

Validated Applications

Western blot 0.25–0.5 µg/mL (Human)
IHC (paraffin sections) 2–5 µg/mL (Human)

Samples with a confirmed band (WB): human HepG2 cells.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:10000, ECL detection.

Samples with confirmed staining (IHC): human liver cancer tissue, human colorectal cancer tissue, human ovarian cancer tissue.

Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.

Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).

Immunogen

Recombinant human Prothrombin fragment (Tyr97–Arg124), expressed in E. coli.

Reactivity Notes

The supplier lists reactivity with human. UniProt describes prothrombin as a secreted protein. Tissue expression noted by UniProt (human): Expressed by the liver and secreted in plasma. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

For research use only; not for diagnostic or therapeutic procedures. Handle with standard laboratory precautions.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 0.25–0.5 µg/mL (Human); IHC (paraffin sections) 2–5 µg/mL (Human). Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human. Other species are not listed by the supplier.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 70.0 kDa; observed band at about 70 kDa.
Q.What immunogen was used to raise this antibody?
A.Recombinant human Prothrombin fragment (Tyr97–Arg124), expressed in E. coli.
Q.How should this antibody be stored?
A.As supplied: −20 °C for up to 12 months from receipt. After reconstitution: 4 °C for up to 1 month, or aliquot and keep at −20 °C for up to 6 months. Avoid repeated freeze–thaw cycles.
