Product Overview
Clone ACBD-6 is a rabbit monoclonal antibody that recognizes prothrombin (gene F2) in human samples and is validated for Western blot and immunoprecipitation. The predicted molecular weight is 70.0 kDa, and the supplier reports an observed band at about 75 kDa.
Prothrombin (F2) is the vitamin K-dependent precursor of thrombin, the central protease of blood coagulation. Thrombin converts fibrinogen to fibrin, activates factors V, VIII, XI and XIII and platelets through PAR receptors, and with thrombomodulin activates the anticoagulant protein C. The prothrombin G20210A variant is a well-known risk factor for venous thrombosis.

Validated Applications
| Western blot | 1:500–1:2000 |
|---|---|
| Immunoprecipitation | 1:20 |
Samples with a confirmed band (WB): human serum.
Recommended loading (WB): 20–40 µg of total protein per lane.
Immunogen
Synthetic peptide based on the sequence of human prothrombin.
Reactivity Notes
The supplier lists reactivity with human. UniProt describes prothrombin as a secreted protein. Tissue expression noted by UniProt (human): Expressed by the liver and secreted in plasma. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
