Product Overview
This rabbit polyclonal antibody targets exopolyphosphatase PRUNE1 in human, mouse and rat samples and is validated for Western blot, ICC/IF, flow cytometry and ELISA. The predicted molecular weight is 50.2 kDa, and the supplier reports an observed band at about 60 kDa.
PRUNE1 is a member of the DHH phosphoesterase family with exopolyphosphatase activity toward short-chain inorganic polyphosphates and phosphodiesterase activity toward cAMP and cGMP. It interacts with cytoskeletal regulators and influences cell migration and proliferation, and biallelic PRUNE1 variants cause a neurodevelopmental disorder with microcephaly.

Validated Applications
| Western blot | 0.25–0.5 µg/mL (Human, Mouse, Rat) |
|---|---|
| Immunocytochemistry / Immunofluorescence | 5 µg/mL (Human) |
| Flow cytometry (fixed cells) | 1–3 µg/1×106 cells (Human) |
| ELISA | 0.1–0.5 µg/mL |
Samples with a confirmed band (WB): human 293T cells, human Jurkat cells, human HEL cells, human MCF-7 cells, rat brain tissue, mouse brain tissue, mouse RAW264.7 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (ICC/IF): U2OS cells.
Samples with a confirmed signal (flow cytometry): 293T cells.
Immunogen
Recombinant human PRUNE1 fragment (Met1–Gln444), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places exopolyphosphatase PRUNE1 in the cytoplasm, nucleus and cell junction. Tissue expression noted by UniProt (human): Ubiquitously expressed. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
