Product Overview
Clone 22P52 is a rabbit monoclonal antibody that recognizes proteasome subunit alpha type-3 (gene PSMA3) in human, mouse and rat samples and is validated for Western blot, IHC, ICC/IF and immunoprecipitation. The predicted molecular weight is 28.4 kDa, and the supplier reports an observed band at about 28 kDa.
PSMA3 (α7) is a non-catalytic α subunit of the 20S core proteasome, forming part of the outer ring that regulates substrate access. It has also been reported to mediate ubiquitin-independent degradation of certain proteins, and it is studied in proteasome biology and protein turnover.

Samples with a confirmed band (WB): human HepG2 cells, human U251 cells, rat liver tissue, rat RH35 cells, mouse liver tissue, mouse HEPA1–6 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 10% SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:500 overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human liver tissue, human liver cancer tissue, human pancreas cancer tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).

The supplier lists reactivity with human, mouse and rat. UniProt places proteasome subunit alpha type-3 in the cytoplasm and nucleus. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
