Product Overview

This rabbit polyclonal antibody detects tyrosine-protein phosphatase non-receptor type 22 (gene Ptpn22) in human, mouse and rat samples and is validated for ELISA, IHC and Western blot. The predicted molecular weight is 89.7 kDa, and the supplier reports an observed band at about 92 kDa.

LYP (PTPN22; PEP in mouse) is a tyrosine phosphatase expressed in immune cells. It limits T-cell receptor signaling by removing activating phosphates from LCK, FYN, ZAP70 and related molecules, and a common PTPN22 variant is a well-known risk factor for several autoimmune diseases.

Validated Applications

Western blot 0.1–0.5 µg/mL
IHC (paraffin sections) 0.5–1 µg/mL
ELISA 0.1–0.5 µg/mL

Samples with a confirmed band (WB): rat thymus tissue, human SW579 cells, human HepG2 cells, human placenta tissue, human HeLa cells, human SW620 cells.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 50 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:10000, ECL detection.

Samples with confirmed staining (IHC): mouse spleen tissue, rat lymphaden tissue.

Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.

Conditions in the example images (IHC): heat-mediated antigen retrieval in citrate buffer (pH 6).

Immunogen

Recombinant mouse PTPN22 fragment (Met1–Gln309), expressed in E. coli.

Reactivity Notes

The supplier lists reactivity with human, mouse and rat. UniProt places tyrosine-protein phosphatase non-receptor type 22 in the cytoplasm. Tissue expression noted by UniProt (mouse): Brain (at protein level). The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 0.1–0.5 µg/mL; IHC (paraffin sections) 0.5–1 µg/mL; ELISA 0.1–0.5 µg/mL. Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human, Mouse, Rat. Other species are not listed by the supplier.
Q.Has this antibody been tested on canine samples?
A.Reactivity with canine samples has not been tested by the supplier. Aligning the immunogen sequence with the canine ortholog (e.g., by BLAST) helps estimate the chance of cross-reactivity.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 89.7 kDa; observed band at about 92 kDa.
Q.What immunogen was used to raise this antibody?
A.Recombinant mouse PTPN22 fragment (Met1–Gln309), expressed in E. coli.
Q.How should this antibody be stored?
A.As supplied: −20 °C for up to 12 months from receipt. After reconstitution: 4 °C for up to 1 month, or aliquot and keep at −20 °C for up to 6 months. Avoid repeated freeze–thaw cycles.
