Product Overview
This rabbit polyclonal antibody targets tyrosine-protein phosphatase non-receptor type 22 (gene PTPN22) in human, mouse and rat samples and is validated for Western blot and ELISA. The predicted molecular weight is 91.7 kDa, and the supplier reports an observed band at about 100 kDa.
LYP (PTPN22; PEP in mouse) is a tyrosine phosphatase expressed in immune cells. It limits T-cell receptor signaling by removing activating phosphates from LCK, FYN, ZAP70 and related molecules, and a common PTPN22 variant is a well-known risk factor for several autoimmune diseases.

Samples with a confirmed band (WB): human A549 cells, human U2OS cells, rat lung tissue, rat PC-12 cells, mouse lung tissue, mouse Neuro-2a cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 50 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.

Recombinant human PTPN22 fragment (Glu207–Thr652), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places tyrosine-protein phosphatase non-receptor type 22 in the cytoplasm. Tissue expression noted by UniProt (human): Expressed in bone marrow, B and T-cells, PBMCs, natural killer cells, monocytes, dendritic cells and neutrophils. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
