Product Overview

Clone 19P55 is a rabbit monoclonal antibody that recognizes pumilio homolog 1 (gene PUM1) in human, mouse and rat samples and is validated for Western blot, IHC, ICC/IF, immunoprecipitation and flow cytometry. The predicted molecular weight is 126.5 kDa, and the supplier reports an observed band at about 140 kDa.

Pumilio homolog 1 (PUM1) is a PUF-family RNA-binding protein that recognizes the UGUANAUA motif in mRNA 3′ UTRs. By recruiting deadenylase complexes and other repressors it lowers the stability and translation of its target mRNAs, with roles in neuronal function, stem cells and fertility.

Synthetic peptide based on the sequence of human pumilio 1.

Reactivity Notes

The supplier lists reactivity with human, mouse and rat. UniProt places pumilio homolog 1 in the cytoplasm and cytoplasmic granule. Tissue expression noted by UniProt (human): Expressed in brain, heart, kidney, muscle, intestine and stomach. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 1:500–1:2000; Immunohistochemistry 1:50–1:200; Immunocytochemistry / Immunofluorescence 1:50–1:200; Flow cytometry 1:60; Immunoprecipitation 1:50. Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human, Mouse, Rat. Other species are not listed by the supplier.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 126.5 kDa; observed band at about 140 kDa. A difference from the calculated size is common and can reflect post-translational modification, processing or isoform differences.
Q.What immunogen was used to raise this antibody?
A.Synthetic peptide based on the sequence of human pumilio 1.
Q.How should this antibody be stored?
A.Long term: −20 °C for up to 12 months. Frequent use: 4 °C for up to 1 month. Avoid repeated freeze–thaw cycles.
