Product Overview
This rabbit polyclonal antibody recognizes pseudouridylate synthase 7 homolog (gene PUS7) in human, mouse and rat samples and is validated for Western blot, IHC, ICC/IF, flow cytometry and ELISA. The predicted molecular weight is 75.0 kDa, and the supplier reports an observed band at about 75 kDa.
PUS7 is a pseudouridine synthase that converts uridine to pseudouridine in tRNAs, mRNAs and small RNAs. Through pseudouridylation of tRNA-derived fragments it helps control protein synthesis in embryonic stem cells, and it is studied in RNA modification biology.

Validated Applications
| Western blot | 0.25–0.5 µg/mL (Human) |
|---|---|
| IHC (paraffin sections) | 2–5 µg/mL (Human, Mouse, Rat) |
| Immunocytochemistry / Immunofluorescence | 5 µg/mL (Human) |
| Flow cytometry (fixed cells) | 1–3 µg /1×106 cells (Human) |
| ELISA | 0.1–0.5 µg/mL |
Samples with a confirmed band (WB): human K562 cells, human MCF-7 cells, human HeLa cells, human 293T cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 10% SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human colon cancer tissuemouse bladder tissue, rat bladder tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Samples with confirmed staining (ICC/IF): A549 cells.
Samples with confirmed staining (ICC/IF): mouse bladder tissue.
Samples with a confirmed signal (flow cytometry): MCF-7 cells.
Immunogen
Recombinant human PUS7 fragment (Asp294–His541), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places pseudouridylate synthase 7 homolog in the nucleus. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
