Product Overview
This rabbit polyclonal antibody recognizes pyrroline-5-carboxylate reductase 1, mitochondrial (gene PYCR1) in human samples and is validated for Western blot, IHC, ICC/IF, flow cytometry and ELISA. The predicted molecular weight is 33.4 kDa, and the supplier reports an observed band at about 35 kDa.
Pyrroline-5-carboxylate reductase 1 (PYCR1) is a mitochondrial enzyme that carries out the final step of proline synthesis, reducing pyrroline-5-carboxylate (P5C) to proline with NAD(P)H. It helps cells cope with oxidative stress, is often upregulated in tumors, and its mutations cause a form of cutis laxa.

Samples with a confirmed band (WB): human SW620 cells, human PANC-1 cells, human 293T cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human lung cancer tissue, human ovarian cancer tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Samples with confirmed staining (ICC/IF): U2OS cells.
Samples with confirmed staining (ICC/IF): human glioma tissue.
Samples with a confirmed signal (flow cytometry): 293T cells.
Immunogen
Recombinant human PYCR1 fragment (Gln10–Asp319), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with human. UniProt places pyrroline-5-carboxylate reductase 1, mitochondrial in the mitochondrion. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
