Overview

This antibody is intended for detection of RANBP2 in biological samples using common immunoassay formats. It is typically selected based on target identity, species reactivity, clonality/clone information, and detection modality.

Vendor notes: Boster Bio Anti-RanBP2 Antibody Picoband® catalog # A00981. Tested in WB applications. This antibody reacts with Human, Mouse, Rat. The brand Picoband indicates this is a premium antibody that guarantees superior quality, high affinity, and strong signals with minimal background in Western blot applications. Only our best-performing antibodies are designated as Picoband, ensuring unmatched performance.

Key elements and design rationale

  • Antibody format: Rabbit Polyclonal Rabbit IgG
  • Immunogen / epitope context: A synthetic peptide corresponding to a sequence at the C-terminus of human RanBP2, different from the related mouse sequence by nine amino acids.
  • Molecular weight context: reported MW: 270 kDa; calculated MW: 358199 MW
  • Reactivity: Human,Mouse,Rat
  • Applications: WB

As a polyclonal antibody, the reagent recognizes multiple epitopes on the target, which can improve detection robustness but may increase sensitivity to sample-dependent epitope changes.

Biological background

E3 SUMO-protein ligase RanBP2. RAN binding protein 2 (RANBP2) is protein which in humans is encoded by the RANBP2 gene. This gene encodes a very large RAN-binding protein that immunolocalizes to the nuclear pore complex. The protein is a giant scaffold and mosaic cyclophilin-related nucleoporin implicated in the Ran-GTPase cycle. And the encoded protein ly interacts with the E2 enzyme UBC9 and strongly enhances SUMO1 transfer from UBC9 to the SUMO1 target SP100. These findings place sumoylation at the cytoplasmic filaments of the nuclear pore complex and suggest that, for some substrates, modification and nuclear import are linked events. This gene is partially duplicated in a gene cluster that lies in a hot spot for recombination on chromosome 2q. Functional note: E3 SUMO-protein ligase which facilitates SUMO1 and SUMO2 conjugation by UBE2I. Involved in transport factor (Ran-GTP, karyopherin)-mediated protein import via the F-G repeat-containing domain which acts as a docking site for substrates. Binds single- stranded RNA (in vitro). May bind DNA. Component of the nuclear export pathway. Specific docking site for the nuclear export factor exportin-1. Sumoylates PML at 'Lys-490' which is essential for the proper assembly of PML-NB. . Reported localization: Nucleus . Nucleus membrane . Nucleus, nuclear pore complex . Detected in diffuse and discrete intranuclear foci (PubMed:11839768). Cytoplasmic filaments (PubMed:7775481). . Expression/tissue context: Higher expression in skeletal muscle and heart and to a lesser extent in liver, brain, placenta, lung, kidney and pancreas.

Research relevance and current trends

  • Cell Cycle: Researchers commonly examine how RANBP2 relates to this theme using model systems and orthogonal readouts.
  • Cell Division: Researchers commonly examine how RANBP2 relates to this theme using model systems and orthogonal readouts.
  • DNA/RNA: Researchers commonly examine how RANBP2 relates to this theme using model systems and orthogonal readouts.

Common research applications

  • Western blotting: compare relative RANBP2 levels across conditions; band patterns may reflect isoforms and processing.

Notes for experimental interpretation

  • Specificity notes: No cross reactivity with other proteins.
  • Cross-reactivity: No cross-reactivity with other proteins.
  • Isoforms and PTMs: Apparent size and signal patterns can differ across splice isoforms, proteolytic processing, and post-translational modifications.
  • Controls: Include an isotype control (as relevant), no-primary control for imaging, and orthogonal validation such as KD/KO samples when available.

We are currently using anti-RanBP2 antibody A00981 for mouse tissue, and we are satisfied with the WB results. The species of reactivity given in the datasheet says human, mouse, rat. Is it likely that the antibody can work on goat tissues as well?
The anti-RanBP2 antibody (A00981) has not been tested for cross reactivity specifically with goat tissues, though there is a good chance of cross reactivity. We have an innovator award program that if you test this antibody and show it works in goat you can get your next antibody for free. Please contact me if I can help you with anything.
