Product Overview
This rabbit polyclonal antibody recognizes ran GTPase-activating protein 1 (gene RANGAP1) in human, mouse and rat samples and is validated for Western blot, IHC, ICC/IF, flow cytometry and ELISA. The predicted molecular weight is 63.5 kDa, and the supplier reports an observed band at about 70 kDa.
RanGAP1 (RANGAP1) is the cytoplasmic GTPase-activating protein for Ran, converting Ran-GTP to Ran-GDP after transport complexes leave the nucleus. This step maintains the Ran gradient that drives nuclear import and export, and RanGAP1 is a classic SUMO-modified protein anchored at the nuclear pore through RanBP2.

Samples with a confirmed band (WB): human SH-SY5Y cells, human 293T cells, human HeLa cells, human A431 cells, human Hacat cells, human MCF-7 cells, human HL-60 cells, human U251 cells, rat brain tissue, rat stomach tissue, rat small intestine tissue, rat pancreas tissue, mouse brain tissue, mouse stomach tissue, mouse small intestine tissue, mouse pancreas tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.25 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human colorectal cancer tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Samples with confirmed staining (ICC/IF): A431 cells.
Samples with a confirmed signal (flow cytometry): U87 cells.

Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places ran GTPase-activating protein 1 in the cytoplasm, nucleus, nucleus envelope and chromosome. Tissue expression noted by UniProt (human): Highly expressed in brain, thymus and testis. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
