Product Overview
This rabbit polyclonal antibody targets regulatory-associated protein of mTOR (gene RPTOR) in human, mouse and rat samples and is validated for Western blot and ELISA. The predicted molecular weight is 149.0 kDa, and the supplier reports an observed band at about 150 kDa.
Raptor (RPTOR) is the defining scaffold subunit of mTOR complex 1 (mTORC1). It recruits substrates such as S6 kinase and 4E-BP1 through their TOS motifs and, with the Rag GTPases, helps bring mTORC1 to the lysosome in response to amino acids, so it is central to studies of nutrient sensing, growth and rapamycin sensitivity.

Samples with a confirmed band (WB): human HepG2 cells, human HeLa cells, human Mcf-7 cells, human Hek293 cells, human A549 cells, rat brain tissue, mouse brain tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.

Recombinant human Raptor/RPTOR fragment (Arg301–Arg358), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places regulatory-associated protein of mTOR in the lysosome membrane, cytoplasm and cytoplasmic granule. Tissue expression noted by UniProt (human): Highly expressed in skeletal muscle, and in a lesser extent in brain, lung, small intestine, kidney and placenta... The samples tested by the supplier (listed above) are a practical starting point for positive controls.
