Product Overview
This rabbit polyclonal antibody targets RAS guanyl-releasing protein 1 (gene RASGRP1) in human samples and is validated for Western blot, flow cytometry and ELISA. The predicted molecular weight is 90.4 kDa, and the supplier reports an observed band at about 100 kDa.
RasGRP1 is a guanine nucleotide exchange factor that switches on Ras in response to diacylglycerol and calcium, then feeds into the ERK/MAP kinase cascade. It couples T-cell and B-cell antigen receptors to Ras and is important for lymphocyte development, and it also shapes NK cell killing and mast cell responses, making it central to immune signaling studies.

Validated Applications
| Western blot | 0.25–0.5 µg/mL (Human) |
|---|---|
| Flow cytometry (fixed cells) | 1–3 µg/1×106 cells (Human) |
| ELISA | 0.1–0.5 µg/mL |
Samples with a confirmed band (WB): human Jurkat cells, human 293T cells, human U-87MG cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with a confirmed signal (flow cytometry): MCF-7 cells, U2OS cells.
Immunogen
Recombinant human RASGRP1 fragment (Arg8–Gln678), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with human. UniProt places RAS guanyl-releasing protein 1 in the cytoplasm, cell membrane, Golgi apparatus membrane and endoplasmic reticulum membrane. Tissue expression noted by UniProt (human): Expressed in brain with higher expression in cerebellum, cerebral cortex and amygdala. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
