Product Overview
Clone AEII-18 is a rabbit monoclonal antibody that recognizes histone-binding protein RBBP4 in human, mouse and rat samples and is validated for Western blot, IHC, ICC/IF, immunoprecipitation and flow cytometry. The predicted molecular weight is 47.7 kDa, and the supplier reports an observed band at about 55 kDa.
RBBP4 (RbAp48) is a WD40-repeat histone-binding protein that helps chromatin regulators reach their histone targets. It is a shared subunit of several complexes, including CAF-1, NuRD, PRC2 and NURF, and so contributes to chromatin assembly after DNA replication, histone deacetylation and gene silencing. The protein is highly conserved.

Samples with a confirmed band (WB): Jurkat cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 10% SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:1000 overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human lymphoma tissue, human glioma tissue, human testis cancer tissue, human bladder cancer tissue, human placenta tissue, human renal cancer tissue, human cervical cancer tissue, human breast cancer tissuehuman lung cancer tissu, human colon cancer tissue, mouse brain tissue, rat brain tissuebr>IP: K562 cells.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Immunogen
Synthetic peptide based on the sequence of human rbAp48.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places histone-binding protein RBBP4 in the nucleus and chromosome. Tissue expression noted by UniProt (human): Expressed in neuroblastoma cells. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
