Product Overview
This rabbit polyclonal antibody recognizes DNA endonuclease RBBP8 in human and mouse samples and is validated for Western blot, IHC, ICC/IF, flow cytometry and ELISA. The predicted molecular weight is 101.9 kDa, and the supplier reports an observed band at about 130 kDa.
RBBP8 (CtIP) is an endonuclease that works with the MRE11–RAD50–NBN (MRN) complex to trim broken DNA ends, the first step of homologous recombination repair. By starting this end resection in S and G2 phase, it steers cells away from non-homologous end joining, and it also works with BRCA1 in DNA damage checkpoints, making it central to DNA repair research.

Samples with a confirmed band (WB): human Raji cells, human A431 cells, human HeLa cells, mouse testis tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human endometrioid adenocarcinoma tissue, human mucinous adenoma of ovary tissue, human spleen tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Samples with confirmed staining (ICC/IF): HeLa cells.
Samples with a confirmed signal (flow cytometry): MCF-7 cells.
Immunogen
Recombinant human RBBP8 fragment (Arg307–Lys702), expressed in E. coli. Sequence identity with the mouse and rat orthologs: 68.9% and 66.7%, respectively.
Reactivity Notes
The supplier lists reactivity with human and mouse. UniProt places DNA endonuclease RBBP8 in the nucleus and chromosome. Tissue expression noted by UniProt (human): Expressed in ER-positive breast cancer lines, but tends to be down-regulated ER-negative cells (at protein level). The samples tested by the supplier (listed above) are a practical starting point for positive controls.
