Product Overview

Clone 23P00 is a rabbit monoclonal antibody that recognizes prohibitin-2 (gene PHB2) in human, mouse and rat samples and is validated for Western blot, IHC and ICC/IF. The predicted molecular weight is 33.3 kDa, and the supplier reports an observed band at about 30 kDa.

Prohibitin 2 (PHB2, also called REA) forms ring complexes with prohibitin 1 in the inner mitochondrial membrane that stabilize respiratory chain components, regulate cardiolipin and OMA1, and act as a receptor for mitophagy. In the nucleus it represses estrogen receptor activity. PHB2 is studied in mitochondrial biology in human and Drosophila.

Reactivity Notes

The supplier lists reactivity with human, mouse and rat. UniProt places prohibitin-2 in the mitochondrion inner membrane, cytoplasm, nucleus and cell membrane. Tissue expression noted by UniProt (human): Expressed in myoblasts. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 1:1000–1:5000; Immunohistochemistry 1:50–1:200; Immunocytochemistry / Immunofluorescence 1:50–1:200. Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human, Mouse, Rat. Other species are not listed by the supplier.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 33.3 kDa; observed band at about 30 kDa.
Q.What immunogen was used to raise this antibody?
A.Synthetic peptide based on the sequence of human REA.
Q.How should this antibody be stored?
A.Long term: −20 °C for up to 12 months. Frequent use: 4 °C for up to 1 month. Avoid repeated freeze–thaw cycles.
