Product Overview
This rabbit polyclonal antibody detects raftlin (gene RFTN1) in human samples and is validated for Western blot, ICC/IF, flow cytometry and ELISA. The predicted molecular weight is 63.1 kDa, and the supplier reports an observed band at about 70 kDa.
Raftlin (RFTN1) is a lipid raft-associated protein involved in membrane trafficking through clathrin and AP-2. In dendritic cells, macrophages and epithelial cells it helps internalize TLR4 after LPS stimulation and delivers poly(I:C) to TLR3-containing endosomes, supporting innate immune signaling, and it also contributes to B-cell receptor signaling.

Validated Applications
| Western blot | 0.25–0.5 µg/mL (Human) |
|---|---|
| Immunocytochemistry / Immunofluorescence | 5 µg/mL (Human) |
| Flow cytometry (fixed cells) | 1–3 µg/1×106 cells (Human) |
| ELISA | 0.1–0.5 µg/mL |
Samples with a confirmed band (WB): human HeLa cells, human Raji cells, human SH-SY5Y cells, human Daudi cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (ICC/IF): U2OS cells.
Samples with a confirmed signal (flow cytometry): U2OS cells.
Immunogen
Recombinant human RFTN1 fragment (Asn18–Lys437), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with human. UniProt places raftlin in the cell membrane, cytoplasm, membrane raft and endosome. Tissue expression noted by UniProt (human): Expressed in B-cells (at protein level). The samples tested by the supplier (listed above) are a practical starting point for positive controls.
