Product Overview
This rabbit polyclonal antibody detects RAD9, HUS1, RAD1-interacting nuclear orphan protein 1 (gene RHNO1) in human samples and is validated for Western blot and flow cytometry. The predicted molecular weight is 26.7 kDa, and the supplier reports an observed band at about 40 kDa.
RHNO1 (also called RHINO) is a nuclear protein that binds the 9-1-1 checkpoint clamp (RAD9–HUS1–RAD1) and helps activate ATR signaling after DNA damage. It also recruits DNA polymerase theta to repair double-strand breaks by microhomology-mediated end joining during mitosis, and it is studied in DNA damage response research.

Validated Applications
| Western blot | 0.25–0.5 µg/mL/ml (Human) |
|---|---|
| Flow cytometry (fixed cells) | 1–3 µg/mL/1×106 cells (Human) |
Samples with a confirmed band (WB): human MOLT-4 cells, human U251 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with a confirmed signal (flow cytometry): RT4 cells.
Immunogen
Synthetic peptide from the N-terminal region of human RHNO1. Sequence identity with the mouse and rat orthologs: 88.2% and 82.4%, respectively.
Reactivity Notes
The supplier lists reactivity with human. UniProt places RAD9, HUS1, RAD1-interacting nuclear orphan protein 1 in the nucleus and chromosome. Tissue expression noted by UniProt (human): Weakly expressed in testis, prostate, ovary, thymus and small intestine. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
