Product Overview
This rabbit polyclonal antibody targets E3 ubiquitin-protein ligase RNF34 in human samples and is validated for Western blot, IHC, ICC/IF, flow cytometry and ELISA. The predicted molecular weight is 41.6 kDa, and the supplier reports an observed band at about 42 kDa.
RNF34 (also called CARP-1 or RIFIFYLIN) is a RING-type E3 ubiquitin ligase with a FYVE domain. It promotes proteasomal degradation of caspase-8 and caspase-10, dampening apoptosis downstream of death receptors, and has also been reported to act on p53 and on proteins in innate immune signaling.

Samples with a confirmed band (WB): human 293T cells, human HEL cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human adrenocortical adenoma tissue, human diffuse large B cells lymphoma tissue, human lung adenocarcinoma tissue, human rectum adenocarcinoma tissue, human testicular seminoma tissue, human tonsil tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Samples with confirmed staining (ICC/IF): A549 cells.
Samples with a confirmed signal (flow cytometry): HEL cells, HL-60 cells.
Immunogen
Recombinant human RNF34 fragment (Glu46–Ser372), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with human. UniProt places E3 ubiquitin-protein ligase RNF34 in the cell membrane, endomembrane system, nucleus and nucleus speckle. Tissue expression noted by UniProt (human): Ubiquitous. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
