Product Overview
This rabbit polyclonal antibody detects mRNA-capping enzyme (gene RNGTT) in human samples and is validated for Western blot, IHC, ICC/IF, flow cytometry and ELISA. The predicted molecular weight is 68.6 kDa, and the supplier reports an observed band at about 69 kDa.
mRNA-capping enzyme (RNGTT) is a two-domain enzyme that carries out the first steps of 5′ cap formation on RNA polymerase II transcripts. Its triphosphatase domain removes the terminal phosphate of nascent mRNA and its guanylyltransferase domain then adds GMP, producing the cap structure that is later methylated and needed for mRNA stability and translation.

Samples with a confirmed band (WB): human Jurkat cells, human HEL cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): Diffuse large B-cells lymphoma of human intestine tissue, human lung adenocarcinoma tissue, human placenta tissue, human rectum adenocarcinoma tissue, human testicular seminoma tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Samples with confirmed staining (ICC/IF): SIHA cells.
Samples with confirmed staining (ICC/IF): human colon cancer tissue.
Samples with a confirmed signal (flow cytometry): SiHa cells.
Immunogen
Recombinant human RNGTT fragment (Leu11–Asp438), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with human. UniProt places mRNA-capping enzyme in the nucleus. Tissue expression noted by UniProt (human): Isoform 1 and isoform 4 (at a lesser extent) are expressed in cerebrum, cerebellum, thyroid, lung, heart, liver, kidney, spleen, large intestine, testis, skin and muscle. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
