Product Overview
This rabbit polyclonal antibody recognizes DNA-directed RNA polymerases I, II, and III subunit RPABC1 (gene POLR2E) in human and rat samples and is validated for Western blot, ICC/IF, flow cytometry and ELISA. The predicted molecular weight is 24.6 kDa, and the supplier reports an observed band at about 23 kDa.
POLR2E (RPB5, RPABC1) is a subunit shared by RNA polymerases I, II and III. In Pol II it forms part of the lower jaw that helps hold incoming template DNA. Because it is common to all three nuclear RNA polymerases, it is studied in general transcription and polymerase assembly.

Validated Applications
| Western blot | 0.25–0.5 µg/mL (Human, Rat) |
|---|---|
| Immunocytochemistry / Immunofluorescence | 5 µg/mL (Human) |
| Flow cytometry (fixed cells) | 1–3 µg/1×106 cells (Human) |

Samples with a confirmed band (WB): human A431 cells, human HeLa cells, human HepG2 cells, human MCF-7 cells, rat liver tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (ICC/IF): U2OS cells.
Samples with a confirmed signal (flow cytometry): HeLa cells.
Immunogen
Recombinant human RPB5/POLR2E fragment (Gln95–Gln210), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with human and rat. UniProt places DNA-directed RNA polymerases I, II, and III subunit RPABC1 in the nucleus. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
