Product Overview

This rabbit polyclonal antibody detects small ribosomal subunit protein eS6 (gene RPS6) in human, mouse and rat samples and is validated for IHC and Western blot. The predicted molecular weight is 28.7 kDa, and the supplier reports an observed band at about 32 kDa.

RPS6 (eS6) is a protein of the 40S ribosomal subunit best known for being phosphorylated at several C-terminal serines by S6 kinases downstream of mTORC1 and by RSK kinases downstream of ERK. Phospho-RPS6 is one of the most widely used readouts of mTOR pathway activity in cell growth and signaling studies.

Validated Applications

IHC (paraffin sections) 0.5–1 µg/mL (Human, Rat)
Western blot 0.1–0.5 µg/mL (Human, Mouse, Rat)

Samples with a confirmed band (WB): rat testis tissue, mouse testis tissue, MCF-7 cells, A549 cells.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 50 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:10000, ECL detection.

Samples with confirmed staining (IHC): rat brain tissue, human intestinal cancer tissue, human lung cancer tissue, rat kidney tissue.

Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.

Conditions in the example images (IHC): heat-mediated antigen retrieval in citrate buffer (pH 6).

Immunogen

Synthetic peptide from the N-terminal region of human RPS6. The peptide sequence is conserved in mouse and rat.

Reactivity Notes

The supplier lists reactivity with human, mouse and rat. UniProt places small ribosomal subunit protein eS6 in the cytoplasm and nucleus. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 0.1–0.5 µg/mL (Human, Mouse, Rat); IHC (paraffin sections) 0.5–1 µg/mL (Human, Rat). Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human, Mouse, Rat. Other species are not listed by the supplier.
Q.Has this antibody been tested on zebrafish samples?
A.Reactivity with zebrafish samples has not been tested by the supplier. Aligning the immunogen sequence with the zebrafish ortholog (e.g., by BLAST) helps estimate the chance of cross-reactivity.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 28.7 kDa; observed band at about 32 kDa. A difference from the calculated size is common and can reflect post-translational modification, processing or isoform differences.
Q.What immunogen was used to raise this antibody?
A.Synthetic peptide from the N-terminal region of human RPS6. The peptide sequence is conserved in mouse and rat.
Q.How should this antibody be stored?
A.As supplied: −20 °C for up to 12 months from receipt. After reconstitution: 4 °C for up to 1 month, or aliquot and keep at −20 °C for up to 6 months. Avoid repeated freeze–thaw cycles.
