Product Overview
Clone AEAE-18 is a rabbit monoclonal antibody that recognizes ribonucleoside-diphosphate reductase subunit M2 (gene RRM2) in human samples and is validated for Western blot, ICC/IF and immunoprecipitation. The predicted molecular weight is 44.9 kDa, and the supplier reports an observed band at about 45 kDa.
RRM2 is the small subunit of ribonucleotide reductase in proliferating cells, providing the tyrosyl radical the enzyme needs to make deoxyribonucleotides. Its levels peak in S phase and control the supply of dNTPs for DNA replication, making it a well-studied target of hydroxyurea and a marker of proliferation.

Validated Applications
| Western blot | 1:500–1:2000 |
|---|---|
| Immunocytochemistry / Immunofluorescence | 1:50–1:200 |
| Immunoprecipitation | 1:50 |
Samples with a confirmed band (WB): human HeLa cells, human SIHA cells, human U251 cells, human MCF-7 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:500 overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Immunogen
Synthetic peptide based on the sequence of human RRM2.
Reactivity Notes
The supplier lists reactivity with human. UniProt places ribonucleoside-diphosphate reductase subunit M2 in the cytoplasm and nucleus. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
