Product Overview
This rabbit polyclonal antibody detects R-spondin-4 (gene RSPO4) in human, mouse and rat samples and is validated for Western blot, flow cytometry and ELISA. The predicted molecular weight is 26.2 kDa, and the supplier reports an observed band at about 26 kDa.
R-spondin-4 (RSPO4) is a secreted member of the R-spondin family that enhances Wnt signaling by binding LGR4–6 receptors and counteracting RNF43/ZNRF3. It is required for nail development, and recessive mutations in RSPO4 cause congenital anonychia.

Samples with a confirmed band (WB): human U2OS cells, human SH-SY5Y cells, human 293T cells, rat brain tissue, mouse brain tissue, mouse Neuro-2a cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with a confirmed signal (flow cytometry): 293T cells.
Immunogen
Recombinant human RSPO4 fragment (Arg60–Lys206), expressed in E. coli. Sequence identity with the mouse ortholog: 82.2%.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt describes R-spondin-4 as a secreted protein. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
