Product Overview
This rabbit polyclonal antibody targets ribosomal protein S6 kinase beta-1 (gene RPS6KB1) in human and mouse samples and is validated for Western blot and flow cytometry. The predicted molecular weight is 59.1 kDa, and the supplier reports an observed band at about 59 kDa.
S6K1 (RPS6KB1, p70S6K) is a serine/threonine kinase activated by mTORC1 in response to growth factors and nutrients. It phosphorylates RPS6, eIF4B and other regulators to promote protein synthesis and cell growth, and it also provides negative feedback to insulin signaling via IRS1; phospho-S6K1 at Thr389 is a standard mTORC1 readout.

Validated Applications
| Western blot | 0.25–0.5 µg/mL (Human, Mouse) |
|---|---|
| Flow cytometry (fixed cells) | 1–3 µg/1×106 cells (Human) |
Samples with a confirmed band (WB): human MOLT4 cells, human U2OS cells, human HL-60 cells, human U251 cells, human 293T cells, human placenta tissue, mouse NIH/3T3 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with a confirmed signal (flow cytometry): MCF-7 cells.
Immunogen
Synthetic peptide from the middle region of human S6K1/RPS6KB1. The peptide sequence is conserved in mouse and rat.
Reactivity Notes
The supplier lists reactivity with human and mouse. UniProt places ribosomal protein S6 kinase beta-1 in the synapse, mitochondrion outer membrane and mitochondrion. Tissue expression noted by UniProt (human): Widely expressed. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
