Product Overview
Clone 3B9 is a mouse monoclonal antibody that recognizes deoxynucleoside triphosphate triphosphohydrolase SAMHD1 in human samples and is validated for Western blot, IHC, ICC/IF and flow cytometry. The predicted molecular weight is 72.2 kDa, and the supplier reports an observed band at about 72 kDa.
SAMHD1 is a dNTP triphosphohydrolase that lowers cellular deoxynucleotide levels, which restricts HIV-1 and other retroviruses in myeloid cells and resting T cells. Phosphorylation switches it toward a separate role in resecting stalled DNA replication forks, and it is studied in antiviral immunity, genome stability and Aicardi-Goutières syndrome.

Samples with a confirmed band (WB): human HeLa cells, human HepG2 cells, human K562 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:10000, ECL detection.
Samples with confirmed staining (IHC): human gastric adenocarcinoma tissue, human ovarian cancer tissue, human breast cancer tissue, human rectal cancer tissue, human gallbladder adenocarcinoma tissue, human lung cancer tissue, human lymphoma tissue, human ovarian serous adenocarcinoma tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Samples with confirmed staining (ICC/IF): Caco-2 cells.
Samples with a confirmed signal (flow cytometry): A431 cells.

Reactivity Notes
The supplier lists reactivity with human. UniProt places deoxynucleoside triphosphate triphosphohydrolase SAMHD1 in the nucleus and chromosome. Tissue expression noted by UniProt (human): Expressed in heart, skeletal muscle, spleen, liver, small intestine, placenta, lung and peripheral blood leukocytes. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
