Product Overview
This rabbit polyclonal antibody detects histone deacetylase complex subunit SAP18 in human, mouse and rat samples and is validated for Western blot, IHC, ICC/IF, flow cytometry and ELISA. The predicted molecular weight is 17.6 kDa, and the supplier reports an observed band at about 18 kDa.
SAP18 is a small protein that joins the SIN3-HDAC corepressor complex and strengthens transcriptional repression. It is also part of the ASAP and PSAP complexes linked to the exon junction complex, where it influences pre-mRNA splicing, and the ASAP complex has been linked to apoptosis.

Samples with a confirmed band (WB): human HeLa cells, human K562 cells, human Jurkat cells, rat RH35 cells, mouse NIN/3T3 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human duodenal adenocarcinoma tissue, human esophageal squamous carcinoma tissue, human liver cancer tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Samples with confirmed staining (ICC/IF): MCF-7 cells.
Samples with confirmed staining (ICC/IF): human esophageal squamous carcinoma tissue, human ovarian cancer tissue.
Samples with a confirmed signal (flow cytometry): U2OS cells.
Immunogen
Recombinant human SAP18 fragment (Met1–Tyr153), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places histone deacetylase complex subunit SAP18 in the nucleus, cytoplasm and nucleus speckle. Tissue expression noted by UniProt (human): Ubiquitous. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
