Product Overview
This rabbit polyclonal antibody detects disks large homolog 1 (gene DLG1) in human, mouse and rat samples and is validated for IHC and Western blot. The predicted molecular weight is 100.5 kDa, and the supplier reports an observed band at about 130 kDa.
DLG1 (SAP97) is a membrane-associated guanylate kinase (MAGUK) scaffold protein that clusters receptors, channels and signaling proteins at specific membrane sites. It supports epithelial polarity and cell junctions and also functions at synapses and the immunological synapse. Its Drosophila counterpart, Discs large, is a classic model tumor suppressor, and DLG1 is studied in cell polarity and signaling.

Samples with a confirmed band (WB): rat lung tissue, mouse lung tissue, HeLa cells, MM231 cells, COLO320 cells, A549 cells, NIH3T3 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human prostatic cancer tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Immunogen
Recombinant human SAP97 fragment (Met1–Ala165), expressed in E. coli. Sequence identity with the mouse and rat orthologs: 89.7% and 84.2%, respectively.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places disks large homolog 1 in the cell membrane, basolateral cell membrane, endoplasmic reticulum membrane and postsynaptic density. Tissue expression noted by UniProt (human): Abundantly expressed in atrial myocardium (at protein level). The samples tested by the supplier (listed above) are a practical starting point for positive controls.
