Product Overview

Clone 20S79 is a rabbit monoclonal antibody that recognizes DNA-binding protein SATB2 in human samples and is validated for Western blot and IHC. The predicted molecular weight is 82.6 kDa, and the supplier reports an observed band at about 90 kDa.

SATB2 is a nuclear matrix-binding transcription factor that organizes chromatin loops and recruits chromatin regulators to control gene expression. It specifies upper-layer cortical neuron identity in the developing brain and drives osteoblast differentiation and craniofacial development, and it is widely used as a colorectal and osteoblastic marker in pathology research.

Validated Applications

Western blot 1:500–1:2000
Immunohistochemistry 1:50–1:200

Samples with a confirmed band (WB): human HT-1080 cells, human HEL cells, human THP-1 cells, human K562 cells.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 10% SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:500 overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.

Immunogen

Synthetic peptide based on the sequence of human SATB2.

Reactivity Notes

The supplier lists reactivity with human. UniProt places DNA-binding protein SATB2 in the nucleus matrix. Tissue expression noted by UniProt (human): High expression in adult brain, moderate expression in fetal brain, and weak expression in adult liver, kidney, and spinal cord and in select brain regions, including amygdala, corpus callosum, caudate nucleus, and hippocampus. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 1:500–1:2000; Immunohistochemistry 1:50–1:200. Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human. Other species are not listed by the supplier.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 82.6 kDa; observed band at about 90 kDa.
Q.What immunogen was used to raise this antibody?
A.Synthetic peptide based on the sequence of human SATB2.
Q.How should this antibody be stored?
A.Long term: −20 °C for up to 12 months. Frequent use: 4 °C for up to 1 month. Avoid repeated freeze–thaw cycles.
