Product Overview
This rabbit polyclonal antibody recognizes succinate dehydrogenase (gene SDHA) in human, mouse and rat samples and is validated for Western blot, ICC/IF and flow cytometry. The predicted molecular weight is 72.7 kDa, and the supplier reports an observed band at about 73 kDa.
SDHA is the flavoprotein subunit of succinate dehydrogenase, the enzyme that is both mitochondrial respiratory complex II and a step in the TCA cycle. It oxidizes succinate to fumarate and passes the electrons on to ubiquinone, and SDHA is studied in mitochondrial disease and as a tumor suppressor in paraganglioma research.

Validated Applications
| Western blot | 0.1–0.25 µg/mL (Human, Mouse, Rat) |
|---|---|
| Immunocytochemistry / Immunofluorescence | 2 µg/mL (Human) |
| Flow cytometry (fixed cells) | 1–3 µg/1×106 cells (Human, Mouse, Rat) |
Samples with a confirmed band (WB): human HeLa cells, human HepG2 cells, human HEK293 cells, human Caco-2 cells, rat brain tissue, rat kidney tissue, rat spleen tissue, rat liver tissue, mouse brain tissue, mouse kidney tissue, mouse liver tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 50 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:100, HRP-conjugated secondary antibody at 1:10000, ECL detection.
Samples with confirmed staining (ICC/IF): HeLa cells.
Samples with a confirmed signal (flow cytometry): HeLa cells, RAW264.7 cells, C6 cells.
Immunogen
Synthetic peptide from the C-terminal region of human SDHA. The peptide sequence is conserved in mouse and rat.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places succinate dehydrogenase in the mitochondrion inner membrane. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
