Product Overview

Clone AADA-19 is a rabbit monoclonal antibody that recognizes succinate dehydrogenase (gene SDHA) in human, mouse and rat samples and is validated for Western blot, IHC, ICC/IF, immunoprecipitation and flow cytometry. The predicted molecular weight is 72.7 kDa, and the supplier reports an observed band at about 73 kDa.

SDHA is the flavoprotein subunit of succinate dehydrogenase, the enzyme that is both mitochondrial respiratory complex II and a step in the TCA cycle. It oxidizes succinate to fumarate and passes the electrons on to ubiquinone, and SDHA is studied in mitochondrial disease and as a tumor suppressor in paraganglioma research.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:500 overnight at 4 °C, HRP-conjugated secondary antibody at 1:500, ECL detection.

Immunogen

Synthetic peptide based on the sequence of human SDHA.

Reactivity Notes

The supplier lists reactivity with human, mouse and rat. UniProt places succinate dehydrogenase in the mitochondrion inner membrane. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
