Product Overview

Clone 24S77 is a rabbit monoclonal antibody that recognizes sentrin-specific protease 2 (gene SENP2) in human, mouse and rat samples and is validated for Western blot, IHC and ICC/IF. The predicted molecular weight is 67.9 kDa, and the supplier reports an observed band at about 68 kDa.

SENP2 is a SUMO-specific protease that both matures SUMO precursors and strips SUMO1, SUMO2 and SUMO3 from target proteins. It shuttles between the nucleus and cytoplasm and is associated with the nuclear pore, and by reversing SUMOylation it influences transcription, protein stability and developmental signaling.

Validated Applications

Western blot 1:500–1:2000
Immunohistochemistry 1:50–1:200
Immunocytochemistry / Immunofluorescence 1:50–1:200

Samples with a confirmed band (WB): human 293T cells, human A431 cells, human U251 cells, human SH-SY5Y cells, rat testis tissue, rat brain tissue, mouse testis tissue, mouse brain tissue.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 10% SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:500 overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.

Immunogen

Synthetic peptide based on the sequence of human SENP2.

Reactivity Notes

The supplier lists reactivity with human, mouse and rat. UniProt places sentrin-specific protease 2 in the nucleus, nuclear membrane, nucleoplasmic side and cytoplasm. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
