Anti-Serotonin Transporter (SERT) (extracellular) Antibody (#AMT-004) is a highly specific antibody directed against an extracellular epitope of the rat protein. The antibody can be used in western blot, immunohistochemistry, immunocytochemistry, indirect flow cytometry, and live cell imaging applications. It has been designed to recognize SERT from human, mouse, and rat samples. Anti-Serotonin Transporter (SERT) (extracellular)-PE Antibody (#AMT-004-PE) is directly conjugated to the Phycoerythrin (PE) fluorophore. This conjugated antibody has been developed to be used in immunofluorescent applications such as direct flow cytometry and live cell imaging.

Target SLC6A4

Overview

Anti-Serotonin Transporter (SERT) (extracellular)-PE Antibody is an antibody targeting Sodium-dependent serotonin transporter, 5-HT transporter, 5-HTT, 5-HTTLPR, SLC6A4 Polyclonal raised in Rabbit (R-Phycoerythrin (R-PE)). This antibody is commonly used in FC, LCI to detect, localize, or compare expression of the target across samples.

Key elements and design rationale

  • Target: Sodium-dependent serotonin transporter, 5-HT transporter, 5-HTT, 5-HTTLPR, SLC6A4 (also reported as Sodium-dependent serotonin transporter, 5-HT transporter, 5-HTT, 5-HTTLPR, SLC6A4).
  • Immunogen/epitope region: 4th extracellular loop.
  • Homology note: Mouse, human - identical (informative for cross-species interpretation).
  • Species reactivity (as provided): Human, Rat, Mouse.
  • Lot quality control (as provided): Western blot analysis (unlabeled antibody, #AMT-004), and direct flow cytometry (labeled antibody)..
  • Peptide confirmation: Confirmed by amino acid analysis and mass spectrometry.
  • Blocking peptide: Available for antigen preadsorption control where appropriate.
  • Conjugate/format: R-Phycoerythrin (R-PE) (may affect detection channel and background).

These attributes help researchers interpret whether signal reflects the intended target in a given assay and sample context.

Biological background

Many physiological, endocrine and behavioral functions are determined and regulated by monoamine signaling1,2. Many brain disorders such as depression, drug abuse, schizophrenia, attention deficit hyperactivity disorder (ADHD) are caused by the malfunction of monoaminergic transmission1-3. The intensity of monoaminergic signaling is determined by the availability of the monoamine, which is in turn determined in part by its uptake from the extracellular milieu via monoamine transporters.

Research relevance and current trends

  • Mapping receptor/channel localization across neuronal subtypes and subcellular compartments.
  • Linking trafficking or surface expression changes to activity-dependent signaling and plasticity.
  • Using KO/KD or blocking-peptide concepts to strengthen antibody-based target assignment.

Common research applications

  • Flow cytometry (direct/indirect): quantify target-positive populations and shifts in expression across subsets.
  • Live cell imaging (LCI): support extracellular-epitope detection on non-permeabilized cells when appropriate.

Interpretation typically benefits from comparing matched sample sets (e.g., treated vs control, WT vs KO/KD) and using orthogonal readouts where feasible.

Notes for experimental interpretation

  • Isoforms and post-translational modifications can shift apparent molecular weight or epitope accessibility across samples.
  • Cross-species signal may depend on epitope conservation; consult the provided homology note when selecting models.
  • Permeabilization, fixation, and antigen retrieval can change accessibility of intracellular vs extracellular epitopes.
  • Conceptual control: antigen preadsorption (blocking peptide) can help assess signal dependence on the immunogen region.
  • Provided control suggestions: Negative control: RIC-001-PE.
  • Application notes: see product-specific dilution/usage notes and control concepts provided in the dataset.
