Product Overview
This rabbit polyclonal antibody targets serpin B9 (gene SERPINB9) in human samples and is validated for Western blot, ICC/IF, flow cytometry and ELISA. The predicted molecular weight is 42.4 kDa, and the supplier reports an observed band at about 38 kDa.
Serpin B9 (SERPINB9, also called PI-9) is an intracellular serpin that inhibits granzyme B, the protease that cytotoxic T cells and NK cells use to trigger apoptosis. By blocking granzyme B it protects killer lymphocytes and other cells from self-inflicted damage and is studied in immune regulation and tumor immune evasion.

Validated Applications
| Western blot | 0.1–0.25 µg/mL (Human) |
|---|---|
| Immunocytochemistry / Immunofluorescence | 5 µg/mL (Human) |
| Flow cytometry (fixed cells) | 1–3 µg/1×106 cells (Human) |

Samples with a confirmed band (WB): human K562 cells, human placenta tissue, human Daudi cells, human Raji cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.25 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (ICC/IF): A549 cells.
Samples with a confirmed signal (flow cytometry): K562 cells.
Immunogen
Recombinant human SERPINB9 fragment (Asp176–Lys313), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with human. UniProt places serpin B9 in the cytoplasm. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
