Product Overview
This rabbit polyclonal antibody detects sestrin-1 (gene SESN1) in human, mouse and rat samples and is validated for Western blot, ICC/IF, flow cytometry and ELISA. The predicted molecular weight is 56.6 kDa, and the supplier reports an observed band at about 56 kDa.
Sestrin-1 (SESN1) is a stress-inducible protein and p53 target that restrains mTORC1 signaling. It binds the GATOR2 complex when leucine is low, and leucine binding releases this block; sestrins also help protect cells against oxidative and genotoxic stress, so SESN1 is studied in nutrient sensing, aging and metabolism.

Samples with a confirmed band (WB): human 293T cells, human K562 cells, rat heart tissue, mouse heart tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (ICC/IF): U2OS cells.
Samples with a confirmed signal (flow cytometry): 293T cells.

Recombinant human SESN1 fragment (Met1–Ala275), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places sestrin-1 in the nucleus and cytoplasm. Tissue expression noted by UniProt (human): Widely expressed. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
